A molecular study of Malassezia strains isolated from cattle with or without otitis was carried out by random amplified polymorphic DNA analysis (RAPD). DNA was extracted and purified from nine strains of Malassezia sympodialis and fourteen of Malassezia furfur. These microorganisms were collected from eight different bovine herds in Minas Gerais state, Brazil. The RAPD analysis and phenograms did not show the formation of genetically distinct groups among the strain isolated from cattle with or without otitis raised in the same herds. Genetic heterogeneity was observed among Malassezia strains from different geographic origins. Molecular characterization of Paecilomyces fumosoroseus (Deuteromycotina: Hyphomycetes) isolates Scientia Agricola

ITS and RAPD analyses were used to investigate molecular variations within samples of Paecilomyces isolates and to resolve five morphologically atypical isolates resembling P. fumosorosus, obtained from whitefly in Northern Paraná State. The ITS4-ITS5 amplicon was 700 base pairs (bp) long in all isolates. The five isolates of Paecilomyces not assigned to species produced restriction profiles identical to all the reference strains of P. fumosoroseus. The extent of fingerprint variability observed by RAPD was sufficient to discriminate all the isolates. ABSTRACT: Swine respiratory diseases such as atrophic rhinitis and bronchopneumonia caused by Pasteurella (P.) multocida cause important economic losses to the modern swine industry. The purpose of this study was to characterize P. multocida strains isolated from swine lungs by RAPD (Randomly Amplified Polymorphic DNA) to demonstrate their genetic diversity. Ninety-four samples of fragments from lungs with pneumonia and sixty one samples without pneumonia were collected in slaughterhouses in Mato Grosso during the period from December 2009 to March 2010. Molecular identification of pollen donor plants on a progeny of Cambona-4 female matrix of maté (Ilex paraguariensis St. Hil. – Aquifoliaceae) - DOI: 10.4025/actascibiolsci.v32i1.764 Biological Sciences

A progeny of Cambona-4, a female maté plant (Ilex paraguariensis) selected by its agronomical characteristics and mild flavor, was evaluated using RAPD markers in order to identify its male parents. Using RAPD markers specifics of each one of the four potential males, the paternity of 84 out of 107 offsprings was confirmed. The majority of the offsprings (83.3%) were ascribed to pollen donor A, while male plants B, C, and D represented 11.9, 4.8, and 0%, respectively. The highly desirable agronomical characteristics and product quality of Cambona-4/pollen donor A offsprings, identified by RAPD markers, lead to the planting of an orchard Cambona-4 and pollinator A to obtain bi-clonal commercial seeds.

Molecular investigation of a fungemia outbreak due to Candida parapsilosis in an intensive care unit BJID

We investigated a nosocomial cluster of four Candida parapsilosis fungemia episodes that occurred in a neurological intensive care unit over a two-week period. The four infected patients had received parenteral nutrition through central lines, and all four had catheter-related candidemia. All of the isolates were susceptible to all of the antifungals tested, including amphotericin B, fluconazole, voriconazole, and caspofungin. For characterization of Xanthomonas species associated with bacterial spot, 161 strains were collected from various field pepper-growing regions. Among them, 131 strains were identified as Xanthomonas euvesicatoria and 30 as Xanthomonas vesicatoria using species-specific primers and polymerase chain reaction followed by restriction fragment length polymorphism analysis. Avaliou-se a variabilidade genética dos estoques de reprodutores e dos peixes jovens de Piaractus mesopotamicus de três pisciculturas do estado do Paraná, utilizadas no programa de aumento de estoque de peixes no rio Paranapanema. Foi utilizado o marcador RAPD para avaliar as amostras do estoque de reprodutores e dos peixes jovens das pisciculturas de Palotina, Cambará e Andirá. A porcentagem de fragmentos polimórficos e o índice de diversidade genética de Shannon dos estoques de reprodutores variaram de 75,0% a 71,4% e de 0,434 a 0,376, respectivamente. Las poblaciones de peces nativos están disminuyendo debido a varios factores que impactan en el ambiente. Como forma de abordar este problema, se han realizado varias acciones como por, ejemplo, el repoblamiento. Sin embargo, estas acciones se realizan sin ningún respaldo científico. Esto puede, además de no ser eficiente, provocar una pérdida de la diversidad genética en las poblaciones, pudiendo ocasionar impactos irreversibles en las próximas generaciones. A México se le considera centro de origen del zapote mamey ya que cuenta con una gran diversidad. Sin embargo, son pocos los trabajos enfocados en estudiar tal diversidad tanto en la parte morfológica, contenido de ácidos graso o aspectos moleculares. Methicillin-resistant Staphylococcus aureus (MRSA) infections frequently complicate the post-operative course of transplant recipients, and despite nasal carriage and endemic colonization, MRSA outbreaks are not commonly described. Acinetobacter baumannii is often implicated in hospital outbreaks in Tunisia. It's a significant opportunistic pathogen associated with serious underlying diseases such as pneumoniae, meningitis and urinary tract infections. The aim of our study was to evaluate its degree of endemicity and its antibiotic resistance evolution essentially in the unit care where its isolation was predominant (57%). This study used 3 methods: antibiotyping, RAPD using 2 primers VIL 1, VIL5 and PFGE with ApaI restriction enzyme. The presence of integron1 and 2 was also studied. Cercospora kikuchii, involved with the defoliation of soybean (Glycine max) plants, is normally associated with Septoria glycines in late season. Seventy-two isolates from different regions in Brazil, obtained mainly from purple stained seeds, showed phenotypic variation. Cercosporin content and rate of colony growth was higly variable among isolates. A strong correlation between cercosporin content and virulence was identified. Genetic variation among and within populations was evaluated based on 86 RAPD loci. The RAPD analysis clustered all isolates into seven groups. Pilocarpine content and molecular diversity in jaborandi Scientia Agricola

Pilocarpine is an imidazol alkaloid exclusively found in Pilocarpus genus and P. microphyllus accumulates its highest content in the leaves. There is no report in the literature on the variability of the pilocarpine content in this genus. A population of 20 genotypes of P. microphyllus from the state of Maranhão, Brazil, was analyzed for Random Amplification of Polymorphic DNA (RAPD) markers and pilocarpine content. Although it was not possible to establish any correlation between these features, the absence or presence of some markers could indicate in some genotypes a possible association with the content of the alkaloid.

Potato cultivar identification using molecular markers PAB

The objective of this work was to evaluate a set of microsatellite markers for varietal identification and characterization of the most widespread potato cultivars in Brazil. The DNA from 14 potato cultivars was genotyped using microsatellite markers and the alleles were scored in silver-stained polyacrylamide gel. Twenty-four microsatellite markers were evaluated, and only one locus was monomorphic. Based on band patterns, a set of two microsatellites that were able to identify and differentiate all examined cultivars was obtained.
